During the 2015 NBA draft, Holmes was selected 2nd round, 37th overall pick by the Philadelphia 76ers of the National Basketball Association(NBA). He signed with the 76ers on July 31, 2015 before he was shipped to play in the summer league for three season.

On November 9, 2015 he made his first career start for his senior side in a loss against the Chicago Bulls. Although they lost, he scored a then season-high 11 points in the game.
