She then appeared as Kheti in the second season of the SABC 2 teen drama Snake Park.

Mbedu landed her first television starring role in the Mzansi Magic teen drama series Is’Thunzi after being unemployed for six months.

She played Winnie in the series, a sassy go-getter who dreams of marrying a wealthy and famous rugby player, only to have her dreams dashed when she is exiled to live with her strict aunt in Bergville. She had a panic attack while filming a rape scene for the show.

In September 2017, she was the only African nominated for an International Emmy Award in the category of Best Performance by an Actress for her role as Winnie in Is’Thunzi.

Mbedu made her international debut in The Underground Railroad, a 2021 Amazon Video historical limited series directed and executive produced by Academy Award winner Barry Jenkins and based on Colson Whitehead’s novel of the same name. Thuso Mbedu received a Hollywood Critics Association Award, an Independent Spirit Award, and a Gotham Award for the series.

It was announced in April 2021 that Mbedu would make her film debut opposite Viola Davis in The Woman King, a historical epic film inspired by true events that occurred in the Kingdom of Dahomey, one of Africa’s most powerful states in the 18th and 19th centuries. Gina Prince-Bythewood will direct from a script written by Fatherhood co-writer Dana Stevens.

She attended Pelham Primary School before continuing her education at Pietermaritzburg Girls’ High School. She later studied Physical Theatre and Performing Arts Management at the University of the Witwatersrand (Wits), graduating with honors in 2013. She had previously studied at the Stella Adler Studio of Acting in New York City in 2012.
